About

The intensive use of antimicrobial agents has facilitated the emergence of a variety of resistance mechanisms in bacteria. The prevalence of resistant bacterial clones is particularly high in the nosocomial environment. The consequences of this problem must be assessed both medically and economically, since the patients infected with antibiotic-resistant microorganisms are in increased risk to receive an inappropriate therapy and to have a longer course of disease or a fatal outcome. Control measures aiming to limit the emergence and dissemination of resistant pathogens include antibiotic restriction policies and development of new antibacterial compounds. The effectiveness of both approaches depends on the good understanding of the biology of resistance. Local monitoring and management is considered essential because of differences in targeting the problems at each place. Moreover, national or global surveillance and strategy develop from local information. As has been documented in recent studies, the resistance rates recorded in Greece are amongst the highest in Europe.

Central activities of the Bacteriology Laboratory of the Hellenic Pasteur Institute (H.P.I.) include basic and applied research in the field of bacterial antibiotic resistance. Our main objectives are the analysis of the mechanisms supporting the development and spread of bacterial resistance and also the surveillance of resistance in the Greek hospitals and the community.

Research projects

(1) Bacterial Resistance to Antibiotics

The project is ongoing since 1992 and up to now has been focused mainly on mechanisms of resistance to the newer β-lactam antibiotics (3rd and 4th generation cephalosporins, β-lactam/β-lactamase inhibitor combinations, and carbapenems) in gram-negative bacilli. Methodological approaches used in the laboratory include: (a) Biochemical characterization of resistance (investigation of the role, structure, and function of bacterial enzymes or other cellular components implicated in the manifestation of resistance). (b) Genetic characterization of resistance (analysis of resistance determinants and their genetic carriers, investigation of the expression of resistance genes). (c) Molecular epidemiology for investigating factors affecting dissemination of resistance among nosocomial pathogens. As an extension of the activities in the field of bacterial resistance, the laboratory is also involved in studies on the evaluation of new antibacterial compounds by measuring their in vitro activity and, more recently, through investigation of their in vivo therapeutic efficacy using experimental infection models in mice.

(2) Neisseria gonorrhoeae – Epidemiology and Antibiotic Resistance

Project  of applied research ongoing since 1982. The project aims to continuous epidemiological surveillance of gonorrhea through demographic and disease markers (incidence, characteristics and behaviour of patients) and microbiological markers, such as types and antibiotic resistance profiles of N. gonorrhoeae strains circulating in the Greek community. The underlying mechanisms of resistance are also investigated for newly recognized or widely disseminated resistance patterns. Due to these activities, the Bacteriology Laboratory of the H.P.I. constitutes, since 1987, the Greek National Reference Centre for Neisseria gonorrhoeae

Activities of the laboratory are supported by the annual budget of the H.P.I. Occasionally, there is complementary external funding from grants and from pharmaceutical companies.

In the evaluation of the Greek Research Institutes and Centres performed by the General Secretariat of Research & Technology (GSRT) in 2005, the Laboratory of Bacteriology was one among three laboratories of the H.P.I. which were evaluated as excellent and funded in the framework of the Excellence GSRT Program.
